Plays important roles in controlling the metabolism of fatty acids at differents levels. Acts as a magnesium-dependent phosphatidate phosphatase enzyme which catalyzes the conversion of phosphatidic acid to diacylglycerol during triglyceride, phosphatidylcholine and phosphatidylethanolamine biosynthesis in the reticulum endoplasmic membrane. Acts also as a nuclear transcriptional coactivator for PPARGC1A/PPARA to modulate lipid metabolism gene expression (By similarity). Is involved in adipocyte differentiation. May also be involved in mitochondrial fission by converting phosphatidic acid to diacylglycerol (By similarity).

Lipin-1 is a Mg(2)+-dependent phosphatidate phosphatase (PAP) enzyme which catalyzes the conversion of phosphatidate to diacylglycerol, a key reaction in glycerolipid biosynthesis (1). It plays critical roles in controlling energy metabolism, has establishing the adipogenic gene expression program during early stages of differentiation, and in triglyceride accumulation in mature adipocytes (2, 3). Lipin-1 also functions as a transcriptional coactivator in conjunction with members of the peroxisome proliferator-activated receptor (PPAR) family (2). There are two isoforms of Lipin-1 (alpha and beta) which appear to serve distinct functions in adipocytes, with unique expression dynamics, subcellular localization, and effects on gene expression. Lipin-1B is the predominant form in mature adipocytes. Lipin-1 expression levels are associated with insulin sensitivity, metabolic rate, hypertension, and risk for the metabolic syndrome (4).
