It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation.

Tumor necrosis factor is a multifunctional proinflammatory cytokine, with effects on lipid metabolism, coagulation, insulin resistance, and endothelial function. Tumor necrosis factor-alpha (TNF-α), the product of activated monocytes, is secreted by macrophages in response to inflammation, infection and cancer (1). TNF-α triggers a signaling cascade, converging on the activation of the transcription factor NF-kappa B, which forms the basis for numerous physiological and pathological processes (2). TNF-α has been shown to play a role in systems such as the pathogenesis of psoriasis, tuberculosis, obesity, cerebral malaria, rheumatoid arthritis and asthma (3).
