Synaptojanin is an inositol 5?0phosphatase and take part in membrane trafficking processes and signal transduction (1). Two isoform of Synaptojanin exists, Synaptojanin 1 (aka Synaptojanin) and Synaptojanin 2. Synaptojanin is mainly expressed in nerve terminals and is involved in clathrin-mediated synaptic vesicle recycling in neuronal cells (2). Ubiquitously expressed and with high catalytic domains homology to Synaptojanin, Synaptojanin 2 is considered as a Rac1 effector. Synaptojanin 2 possibly medicates the inhibitory effect of Rac1 on clathrin-mediated endocytosis. Synaptojanin 2 contributes to Rac1?s role in cell invasions and migration by regulation the formation of specialized membrane structures in ECM (invaodopoida and lamellipoida). Synaptojanin 2 is considered to be a novel target for therapeutic intervention in malignant tumors.
