Tumor necrosis factor receptor superfamily member 4 (OX40, CD134) is a membrane receptor found on activated CD4+ and CD8+ T cells and B cells (1). OX40 binds to OX40 ligand (OX40L) and other TNF receptor-associated factors (TNAF2, TRAF3, TRAF5). OX40-OX40L complex produces CD28 independent co-stimulatory signals. OX40 is involved in the pathologic cytokine storm associated in certain viral infection, such as H4N1 bird flu. OX40 starts a survival signal to the T-cell preventing activation-induced apoptosis, but up regulates cytokine production. Inhibition of OX40 using OX40-immunoglobulin has been linked to reduction of immune overreaction without affecting the viral clearance (2).
