Adapter protein that functions as clathrin-associated sorting protein (CLASP) required for clathrin-mediated endocytosis of selected cargo proteins. Can bind and assemble clathrin, and binds simultaneously to phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) and cargos containg non-phosphorylated NPXY internalization motifs, such as the LDL receptor, to recruit them to clathrin-coated pits. Can function in clathrin-mediated endocytosis independently of the AP-2 complex. Dab2 (Disabled homolog 2, Diabled-2) is a mammalian structural homolog of Drosophila Disabled protein and a mitogen-responsive phosphoprotein (1). As a component of the CSF1 signal transduction pathway, Dab2 is normally expressed in ovarian epithelia cells. However, in ovarian carcinoma, Dab2 expression is either lost or down-regulated. Therefore, Dab2 is speculated to be a negative regulator of growth and its down-regulation considered to play critical role in ovarian carcinogenesis (2).
