Protein phosphatase 2C (PP2C) is a family of serine/threonine specific protein phosphatases. PP2C is a 42 kDa monomeric enzyme with broad substrate specificity and its activity is depended upon divalent cations (e.g. magnesium) (1). In mammals, three members of PP2C family are known; PP2C -alpha, -beta, and -gamma. PP2C-alpha functions by dephosphorylating various substrates (e.g. Smad1, MAPK) and it is considered to be a negative regulator of cell stress pathway (2). PP2C-alpha has been implicated in the regulation of apoptosis-, p53-, cyclin-dependent kinase-, and TGF beta- signaling. Studies have suggested tumor-suppressing properties of PP2C -alpha and its possible role in cancer development and progression (3).

Function
Enzyme with a broad specificity. Negatively regulates TGF-beta signaling through dephosphorylating SMAD2 and SMAD3, resulting in their dissociation from SMAD4, nuclear export of the SMADs and termination of the TGF-beta-mediated signaling. Dephosphorylates PRKAA1 and PRKAA2. Plays an important role in the termination of TNF-alpha-mediated NF-kappa-B activation through dephosphorylating and inactivating IKBKB/IKKB.

Nucleus. Cytoplasm, cytosol. Membrane. Note=Weakly associates at the membrane and N- myristoylation mediates the membrane localization.
